1. Lethal dose 50 of Mahogany seeds:
a. Mahogany seed 100% decoction
The mice manifested decrease motor activity, watery stool, decrease rate and depth of respiration, loss of screen grip, and convulsion then death. No gross morphological differences from the control group was observed.
The lethal dose 50 of mahogany seed 100% decoction is 147.66g/kg
b. Mahogany seeds neutralized
The mice manifested decrease motor activity, watery stool, decrease rate and depth of respiration, loss of screen grip, and convulsion, Death follows after 30 minutes to one hour of administration. No gross morphological differences from the control group was observed.
The neutralized extract of mahogany seeds has an LD50,of 118.23g/kg.
c. Mahogany seed alcoholic extract
The mice showed the same manifestations as the decoction and neutralized extract mahogany seeds.
The lethal dose 50 of mahogany seed alcoholic extract is 31.96g/kg which is more potent than the decoction and neutralized extract.
d. Mahogany Lipid extract
At a dose of 10g/kg mice manifested increased urination, decreased motor activity, watery stool, decrease rate and depth respiration, pilomotor erection, loss of screen grip, loss of writhing reflex, convulsion, then death. At a lowest dose of 10g/kg mice died after two days of drug administration.
Lethal dose 50 studies show that mahogany lipid extract 10g/kg is the most lethal preparation followed by mahogany seed alcoholic extrcat which has an LD50 of 31.46g/kg; followed by mahogany extract with an LD50 of 118.23g/kg; followed by 100% decoction which has an LD50 of 147.66 g/kg.
2. Behavioral Studies:
a. Pole jumping
Results show a delay of responding to the conditioned stimulus (bell) an average of 15 seconds.
b. Spontaneous Motor Activity was also depressed.
The LD50 studies show that mahogany lipid extract is the most potent preparation. Unless the preparation of this is saddled with difficulty and expense, the lipid extract is preferred for further pharmacologic studies. The behavioral studies while lending support to a tranquilizing activity, the toxidrome results to a point more towards a general depression of the CNS.
Further studies in behavior should be postponed to give preference for screening studies in other organ systems. This section could screen for smooth muscle effects on the trachea, heart muscle studies, uterus (pregnant, nonpregnant) and blood vessels.
